About Mounjaro 5 mg
Mounjaro (tirzepatide) 5 mg is a prescription medication administered as a once-weekly subcutaneous injection to help manage type 2 diabetes in adults. It is also increasingly recognized for its role in weight loss, though this use may be off-label in some regions. Manufactured by Eli Lilly and Company, Mounjaro is part of a class of drugs that target both glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors, offering a dual mechanism to regulate blood sugar and support weight management.

Use for Type 2 Diabetes

Mounjaro 5 mg is FDA-approved for adults with type 2 diabetes to improve blood sugar control alongside diet and exercise. It works by mimicking GIP and GLP-1 hormones, which enhance insulin production when blood sugar levels are high, reduce glucose production by the liver, and slow digestion to prevent rapid blood sugar spikes. In clinical studies, 75% to 90% of patients achieved an A1C of less than 7% with Mounjaro doses ranging from 5 mg to 15 mg, starting from an average A1C of 7.9% to 8.6%. The 5 mg dose is typically the first maintenance dose after a 4-week initiation period at 2.5 mg, with potential increases in 2.5 mg increments every 4 weeks if additional glycemic control is needed, up to a maximum of 15 mg weekly. It can take 8 to 12 weeks to reach target A1C levels, with significant improvements often seen by 40 to 52 weeks. Mounjaro is not suitable for type 1 diabetes or children under 18, and its safety in patients with a history of pancreatitis is unknown.

In a 2024 study, 81.8% of adults with overweight or obesity and type 2 diabetes achieved at least 5% weight loss within one year using Mounjaro, compared to 66.5% with Ozempic. Weight loss can begin as early as 28 weeks, but individual results vary.

Warnings and Contraindications

Mounjaro carries a boxed warning for the risk of thyroid C-cell tumors, including medullary thyroid carcinoma (MTC), based on animal studies. It is contraindicated in patients with a personal or family history of MTC or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Serious side effects include pancreatitis (severe abdominal pain, with or without vomiting), hypoglycemia (especially when used with insulin or sulfonylureas, causing dizziness, sweating, or confusion), acute kidney injury (due to dehydration from nausea, vomiting, or diarrhea), and gallbladder problems (e.g., gallstones, with symptoms like upper abdominal pain or jaundice).
